Is there any difference in connotation between 大きい and 大き? Or are they just synonyms that sound almost the same?
大きい and 小さい are (I think) the only two adjectives that appear as both い-adjectives and な-adjectives. おおき(な) is just the な-adjective form of おおきい.
They are technically attributive modifiers (連体詞), not true な-adjectives
Basically, they can't be used as predicates.
静かな部屋 - 静かです The な gets dropped when used as a predicate (typical for true な-adjectives).
大きな車 - 大きです - Doesn’t work. You have to use 大きい instead.
大き (noun | な-adj) = a large amount, a great deal, is it's own different thing altogether.

I think this この違いは大きです。technically works (grammatically), but it would be very weird in everyday speech. 大き is archaic/literary.
